PloS one, 12(9), e0183315. In this study, a CHO DG44 cell line was adapted to to serum free conditions, transfected with CR9114L expressing plasmids. A healthy, stable clone, 15G6, was selected, that shows robust antibody production. Antibody produced by this clone showed activity in binding and functional assays comparable to mAb produced by transient expression in 293F cells. The FBS weaning process of CHO DG44 cells was tested in DMEM/Ham's F12 medium, ProCHO5, CD DG44, OptiCHO, and CHO-S-SFM II media. The specific growth rate and cell viability were observed and CHO-S-SFM II medium was determined as the best option for cultivating this newly adapted CHO DG44 serum-free cell line.
