Kim, M. S., Yun, J. I., Gong, S. P., Ahn, J. Y., Lim, J. M., Song, Y. H., ... & Lee, S. T. (2015). Development of a chemically defined in vitro culture system to effectively stimulate the proliferation of adult human dermal fibroblasts. Experimental dermatology, 24(7), 543-545. In this study, a serum-free CDCS that stimulates proliferation and inhibits senescence of aHDFs cultured in vitro was established by replacing FBS with KSR, providing extracellular signals derived from the ECM protein FN and re-optimizing the concentration of each culture component. The optimized CDCS increased the proliferation of aHDFs significantly, without any increase in the senescence rate or alteration in the morphology of aHDFs, despite long-term culture. A detailed description of how the media was developed is present. https://onlinelibrary.wiley.com/doi/pdf/10.1111/exd.12695 |
