Felka, T., Schäfer, R., De Zwart, P., & Aicher, W. K. (2010). Animal serum-free expansion and differentiation of human mesenchymal stromal cells. Cytotherapy, 12(2), 143-153. In this study, FCS-free media were tested for the expansion and differentiation of MSC in compliance with the European good manufacturing practice (GMP) regulations for medicinal products. MSC expansion medium was modified by replacing FCS with human plasma and platelet extract. The analysis of MSC expanded in FFPP medium showed a high proliferation capacity, with preservation of the ability of the cells to differentiate into adipogenic, osteogenic and chondrogenic lineages. Human plasma is a suitable FCS replacement for the expansion and differentiation of MSC, providing a feasible alternative for tissue engineering with GMP-compatible protocols. The formulation of FFPP medium is listed in the Methods, under the heading; Isolation of hMSC and cell culture. https://ac.els-cdn.com/S1465324910703765/1-s2.0-S1465324910703765-main.pdf?_tid=b93c1757-53c5-4838-88c5-59be0fd794e3&acdnat=1534145220_982242f56eb94a3d9007b8e1e1f3e26c |
