Stem cell research, 6(1), 34-49. This study presents a novel, single-step method of generating murine ESC-derived chondrocytes in monolayer cultures under chemically defined conditions. Supplementation of ESC cultures with BMP4 (10 ng/ ml), activin A (30 ng/ml), or Wnt3a (100 ng/ml) on Day 0 of differentiation (dark blue thunderbolt) successfully induced mesoderm progenitors. N2B27 SF medium was supplemented with different components as specified in the Materials and Methods, under the heading; ESC maintenance and differentiation cultures.
