Lee, M. O., Jeon, H., Son, M. Y., Lee, S. C., & Cho, Y. S. (2017). Clump-passaging-based efficient 3D culture of human pluripotent stem cells under chemically defined conditions. Biochemical and biophysical research communications, 493(1), 723-730. In this study, a novel, simple clump-passaging method for in vitro hPSCs 3-dimensional (3D) culture was developed that can be exploited for large-scale production. Fully grown hPSC spheroids were dissociated into smaller-sized spheroid clumps by simple treatment with enzyme-free dissociation buffer, and clumped hPSCs were inoculated and maintained for 3D suspension culture. Chemically defined media formulations that are suitable for 3D culture and commercially available were tested. Defined medium suitable for clump-passaging-based 3D culture of hPSCs. L7 media ,SFM media, and TeSR-E8 media showed relatively high growth rates, whereas the mTeSR-1 media, PluriSTEM media and STEMium media showed relatively lower growth rates, respectively. L7 media , SFM media, and TeSR-E8 media are commercial products. Their formulation is proprietary. https://ac.els-cdn.com/S0006291X17317072/1-s2.0-S0006291X17317072-main.pdf?_tid=fe1de013-2b2f-4c24-a661-9f77f8b0a0c1&acdnat=1530019742_777d1f5b41d0f1fd0a94abbbb914b41f |
