Multiple genotypic aberrances associate to terminal differentiation‐deficiency of an oral squamous cell carcinoma in serum‐free culture. Differentiation, 76(8), 868-880. In this study, an oral squamous cell carcinoma (OSCC) cell line was established in the serum-free condition optimized for normal oral keratinocytes (NOK), and use of NOK as comparator aided the precise definition of the transformed state. Keratinocyte-SFM or the laboratory-fabricated variant termed EMHA (epithelial medium with high levels of amino acids, Grafström, 2002), both of which contained growth-promoting supplements like epidermal growth factor and pituitary extract, were used interchangeably in these studies without detectable differences in growth among cell lines or outcome of experiments. The resultant LK0412 line represented an immortal and malignant tumor variant with preserved characteristics of the original tumor. Keratinocyte-SFM is a commercially available product. The precise formulation is proprietary. EMHA is the laboratory-fabricated variant. The formulation of EMHA is listed in Grafström, 2002. |
