Arthuso, F. S., Bartolini, P., & Soares, C. R. J. (2012). Laboratory production of human prolactin from CHO cells adapted to serum-free suspension culture. Applied biochemistry and biotechnology, 167(8), 2212-2224. This study describes a fast and practical laboratory adaptation of Chinese hamster ovary (CHO) cells transfected with a eukaryotic hPRL expression vector, in ~40 days, to grow in suspension in serum-free medium. CHO-pEDdc-hPRL cells were adapted to serum-free suspension culture by a three-step procedure. For laboratory production, the cells were cultured in CHO-S-SFM II medium, supplemented with antibiotics, as specified in the Materials and Methods, under the heading; Lab Scale Production in Spinners. CHO-S-SFM II medium is a commercially available product. The precise formulation is proprietary. https://link.springer.com/content/pdf/10.1007%2Fs12010-012-9745-1.pdf |
