An XBP-1 dependent bottle-neck in production of IgG subtype antibodies in chemically defined serum-free Chinese hamster ovary (CHO) fed-batch processes. Journal of biotechnology, 135(2), 217-223. In this study, using CHO cells grown in suspension in chemically defined serum-free media, we have assessed the possibility to improve fed-batch processes by stable expression of the active spliced form of human XBP-1. We show that in inoculum suspension cultures this approach can lead to an increase in ER-specific antibody productivity and final process yield without altering product quality of an IgG therapeutic. Suspension cultures of monoclonal antibody (mAb) producing CHO-DG44 cells and stable transfectants thereof were incubated in BI proprietary chemically defined, serum-free medium.
