In this study is shown that both human adult corneal stromal (SSC) and epithelial limbal (LSC) stem cells can be isolated from small superficial limbal biopsies and primary cultured in the same feeder-free and xeno-free culture conditions. Isolated cells were cultured with Essential 8 medium (E8), E8 supplemented with EGF (E8+) or Green's medium with 3T3 feeder-layers. The highest number of spheres was obtained with ILC-scraping+ grown in E8. These cultured stem cells feature high growth potential and the expected phenotypes and differentiation potentials in primary culture.
