Chang, S. H., Chiang, I. N., Chen, Y. H., & Young, T. H. (2013). Serum-free culture of rat proximal tubule cells with enhanced function on chitosan. Acta biomaterialia, 9(11), 8942-8951. The purpose of this study is to culture rat renal proximal tubule cells (PTCs) on chitosan without serum to maintain a transcellular pathway to transport water and ions effectively without loss of highly differentiated cell function. In this study, a successful primary culture of PTCs has now been produced on chitosan in serum-free culture condition, which offers potential applications for chitosan in renal tissue engineering. The effect of chitosan, which is structurally similar to glycosaminoglycans, in the absence of serum on the primary cultured PTCs was compared that of collagen with or without serum. The culture medium consists of equal parts of DMEM/F12 and keratinocyte serum-free medium supplemented with different components as specified in the Materials and Methods, under the heading; 2.2. Isolation of rat renal proximal tubules. https://ac.els-cdn.com/S1742706113003255/1-s2.0-S1742706113003255-main.pdf?_tid=4977e7f9-fdd9-46a2-bbce-4ea73089d75b&acdnat=1533305995_e0a1d471a4d5987285bf5d0dc565299d |
