In this work, a scalable system for HDAd production is designed and evaluated focusing on the co-infection step. A human embryo kidney 293 (293) derived cell line, the 293SF/FLPe was generated to produce efficiently HDAd while restricting the packaging of HAd. The best FLPe clone was adapted to grow in serum-free NSFM13 supplemented with 0.75 µg/mL of puromycin. This new cell line is referred to as 293SF/FLPe cell line.

Serum-free NSFM13 medium was supplemented with different components as specified in the Materials and Methods, under the heading; 2.2. Cell lines and culture media. The preperation of NSFM13m medium is described in a previous article, Nadeau et al. 2002.
