Sato, Y., Wakitani, S., & Takagi, M. (2013). Xeno-free and shrinkage-free preparation of scaffold-free cartilage-like disc-shaped cell sheet using human bone marrow mesenchymal stem cells. Journal of bioscience and bioengineering, 116(6), 734-739. In this study, the xeno-free cultivation method to obtain a scaffold-free cartilage-like disc-shaped cell sheet using mesenchymal stem cells (MSCs) derived from human bone marrow without the shrinkage of the sheet was investigated. MSCs were inoculated into Cell Culture Insert using serum-free chondrogenic differentiation medium containing TGF-beta3, IGF-1 and dexamethasone or other modified media, and cultured at 37 degrees C in 5% CO2 for 3 weeks. The scaffold-free and xeno-free cartilage-like cell sheet formed by the cultivation of human bone marrow MSCs in chondrogenic differentiation medium containing human serum showed no shrinkage. The formulation of chondrogenic differentiation medium is listed in the Materials and Methods, under the heading; Chondrogenic differentiation. https://ac.els-cdn.com/S1389172313002016/1-s2.0-S1389172313002016-main.pdf?_tid=6fbf1b4f-1181-40f7-9f35-a6faf1e83d41&acdnat=1533215668_a708f447c14fe2a7eebee8a427470237 |
